Yeast experts

Hi fellow rats,

I received a S. Cerevisiae strain that doesn't express Ura3. I assume this yeast needs supplemented uracil to grow. I received a stab and I want to grow it out. Should I just use ypd agar supplemented with uracil or does it need additional nutrients?

I would like to express a secreted protein in this yeast. Should I clone my orf after the KR amino acid sequence or after the KREAEA sequence. I'm generally familiar with mammalian secretion tags but not yeast. I tried to use signalp to show me the correct cleavage site but it doesn't do yeast well. Moreover, I've heard both locations are correct from various sources. Who is right? Ideally, I don't want extra amino acids in the n terminal portion of my protein.

Thank you all.
