dendritic cells

I work with dendritic cells, and I'm about to start an infection experiment, with the goal of extracting RNA. This is my first time with these cells and I have a question about their replication. I would like to activate them on Tuesday, but since the medium will already be exhausted, I would need to replication them on Monday. But I'm afraid they're still in the form of monocytes. I'd like some tips.
