Advice/info on depression & dosing

Hi, my friend suffers from depression. He has tried several times to “break through” but can’t seem to make it happen. He has tried brews, smoking, and pharmahuasca, even all three together. Now he’s wondering if the abnormality of his serotonergic pathway (related to depression) is preventing him from getting there. Just trying to get some input.
