Help with chaperone/protein coexpression
Hello labrats, I'm facing some issues with coexpressing a protein and it's chaperone (both meant to localise to the periplasm via their sec signal peptides) with the pETDuet system. So long story short, from my westerns I can see a nice clean band for the chaperone in the periplasmoc fraction (extracted with osmotic shock) but the associated protein is nowhere to be seen.
My fear is that it's gone into inclusion bodies - anyone have experience with coexpressing two proteins on the same plasmid that end up being localised to different locations?
