While David Sinclair’s Information Theory of Aging suggests that resetting a cell's "identity" via the epigenome acts as a master switch for rejuvenation, credible critics argue this "silver bullet" approach fails to address physical, indigestible waste that the human body simply never evolved to clear. Even if a cell is epigenetically "reprogrammed" to a youthful state, it remains physically trapped by metabolic byproducts like lipofuscin, an indigestible "aging pigment" that accumulates in long-lived cells like neurons and heart muscle. This "garbage catastrophe" occurs because lipofuscin is chemically resistant to lysosomal degradation; a cell that "identifies" as young still has its internal machinery physically clogged by these aggregates, which inhibit vital functions regardless of the cell's genetic signaling.

The limitation of epigenetic reprogramming is even more pronounced with 7-keto-cholesterol (7KC) and glucosepane, which represent damage that is often beyond the cell's reach.

 * 7-Keto-cholesterol: This toxic oxysterol is a major driver of arterial plaques and is highly cytotoxic, disrupting cell membranes and inducing cell death. Because it is a non-enzymatic byproduct of cholesterol oxidation, the body lacks a robust intrinsic mechanism to safely "re-metabolize" it once it has reached high concentrations in the artery walls.

 * Glucosepane: As a primary advanced glycation end-product (AGE), glucosepane forms "crosslinks" that physically glue extracellular proteins like collagen together, causing the stiffening of arteries and skin. Since these bonds are extracellular, resetting the "software" inside the cell does not provide a mechanism to snap the chemical bonds outside the cell.

Ultimately, these examples illustrate a "hardware" problem that "software" updates cannot fix. Resetting a cell's identity may restore some youthful repair signaling, but it cannot grant the cell new "tools"—such as specialized enzymes—that are not already present in the human genome. For metabolic "dead ends" like lipofuscin and glucosepane, proponents of the SENS framework argue that we must supplement the body’s natural abilities with external, engineered biotechnologies rather than relying solely on the body's intrinsic, and currently insufficient, repair systems.

We need a cure for cancer, but that is far from good enough to add decades to the average human lifespan. And I'm betting a perfect epigenome fix isn't close to good enough either, despite what some would have us think.
