What do you change first after bolting on a new 4-barrel carb?

I am planning a carb swap using Evil Energy fuel system parts and want to shorten the tuning process as much as possible. After the install, I usually see people start with idle mixture, timing, and jetting, but I want to hear what actually made the biggest difference on your build. What was your first adjustment?
