ELI5: tilesets and spritesheets

When one finds a spritesheet, which is lots of (let's say) 16x16 characters to be "cut up", how does one do that? Cut and save individually with MS Paint or something?

With respect to both, how are tilesets and sprites actually used? Are they individual .png .jpegs or something?
