Borderlands 2 Questions

I've seen people do it to where there 2 instances of the game running and one person playing with controller and another with mouse and keyboard, is it possible to do that and play online with some other people. For instance I play mouse and keyboard on my monitor and then my gf plays on the tv with her controller while we play with some of my buds online. Thanks for the help in Advance :)
